Am Ende des ersten Films wurde der mächtige dunkle Zauberer Gellert Grindelwald (Johnny Depp) mit Hilfe von Newt Scamander (Eddie Redmayne) vom MACUSA (Magischer Kongress der Vereinigten Staaten von Amerika) gefangen genommen. Doch Grindelwald machte seine Drohung wahr, entkam der Haft und machte sich daran, Anhänger zu sammeln, ohne von seiner wahren Absicht zu ahnen: reinblütige Zauberer zu erziehen, um über alle nichtmagischen Wesen zu herrschen. Um Grindelwalds Pläne zu vereiteln, engagiert Albus Dumbledore (Jude Law) seinen ehemaligen Schüler Newt Scamander, der sich bereit erklärt, zu helfen, ohne sich der Gefahren bewusst zu sein, die vor ihm liegen.
